Survivin and Mucin 2MUC2 Detection in Iraqi Patients with Colorectal Tumors

Abstract
Survivin is the smallest member of the inhibitors of apoptosis protein (IAPs) family. Survivin's overexpression blocks both intrinsic and extrinsic apoptosis pathways and it is highly expressed in most cancers and malignancies. Mucin 2MUC2 is the major gel forming mucin in the colon and the first human secretory mucin to be identified and characterized. To analyze the association between survivin, mucin 2 (MUC2) and the clinicopathological features on tissues from a group of patients with colorectal tumors in mid-Euphrates-Iraq. One-hundred twenty formalin-fixed, paraffin- embedded colorectal tissues enrolled in this study; 60 colorectal tissue biopsies from malignant colorectal tumors; 40 tissues obtained from patients with benign colorectal tumors and 20 apparently healthy colorectal tissues were used as control group. Detection of mutated survivin gene was done by chromogenic in situ ybridization (CISH) technique while the protein expression of MUC2 by using immunohistochemistry (IHC) detection system. The positive results of mutated survivin gene -CISH detection in malignant and benign colorectal tumors, were 63.3%, 55%, respectively. The present study shows the positive results of MUC2-IHC detection in malignant, benign and apparently healthy group colorectal tissue, were 50.0%, 25.0% and 20.0%, respectively. The significant co-expression of mutated expression of survivin gene as well as MUC2 in colorectal tissues could point for their possible role in either colorectal pathogenesis or carcinogenesis.
